Lipid Test Cassettes
The following lipid cassettes are available for use with the L·D·X®:
The Cholestech L·D·X® system combines enzymatic methodology with solid phase technology to measure total cholesterol, HDL cholesterol, triglycerides and glucose. Samples for testing can be whole blood from a fingerstick (collected in lithium heparin coated capillary tubes) or venipuncture. A unique system in the cassette separates the plasma from the blood cells. A portion of the plasma flows to the right side of the cassette and is transferred to the total cholesterol and triglyceride reaction pads; Simultaneously, plasma flows to the left side of the cassette where the low density lipoproteins (LDL and VLDL) are precipitated with dextran sulphate and magnesium acetate precipitating agent. The filtrate, containing both HDL and glucose is transferred to those reaction pads. Cholesterol esterase hydrolyses the cholesterol esters to free cholesterol and fatty acid; cholesterol oxidase then oxidises free cholesterol; the resulting peroxide reacts with TOOS (4-aminoantipyrine and N-ethyl-N-sulfohydroxypropyl-m-toluidine, sodium salt) in a reaction catalysed by horseradish peroxidase, to form a purple coloured quinoneimine dye - proportional to the TC and HDL cholesterol concentrations in the sample. In a similar way the L·D·X® measures the glucose concentration by an enzymatic method that uses glucose oxidase to catalyse the oxidation of glucose to peroxide. Triglycerides concentration is measured via an enzymatic method based on the hydrolysis of triglycerides by cholesterol esterase to glycerol and free fatty acids. Glycerol, in a reaction catalysed by glycerol kinase is converted to glycerol-3-phosphate, which is in turn oxidised to peroxide. The colour reaction is the same as for TC, HDL and glucose. The brown, magnetic stripe on each cassette contains the calibration information required for the L·D·X® analyser to convert the reflectance reading to the total cholesterol , HDL cholesterol, triglycerides and glucose concentrations.
